Abstract
(Human Genetics and Genomics Advances 5, 100306; July 18, 2024) The in vitro model for functionally testing PMS2 variants of undetermined significance (VUSs) was mistakenly enriched via selection with G418 instead of puromycin. This experiment has been repeated in its entirety from the point of lentiviral infection with the following modification: (1) cells were selected with and grown continuously in puromycin, (2) a lower dose of 6-TG was used for the mismatch repair (MMR) assay, and (3) doxycycline was spiked into the MMR experiment daily to ensure continuous expression of PMS2 from the transduced lentiviral cassette. Figures 6C and 6D have been updated to reflect these changes, as have the corresponding methods and results sections. Although these updates do not change the conclusions of the paper, the authors regret the error.
